Brief Definitive Report PARALLEL SYNTHESIS OF IMMUNOGLOBULINS AND J CHAIN IN POKEWEED MITOGEN-STIMULATED NORMAL CELLS AND IN LYMPHOBLASTOID CELL LINES* BY JIRI MESTECKY,

چکیده
In sera and in secretions, J chains are found linked to molecules of polymeric IgA and IgM by disulfide bonds. Their function remains undetermined; the init ial concept that they were essential to the process of polymerization has been questioned. It is of special interest that J-chain synthesis is not restricted to cells producing polymeric immunoglobulins of the IgA and IgM classes; it has been detected in murine plasma cells that produce IgG as well as in those that synthesize only L chains (1-4). Within such cells, J chain was thought to be either linked to a small portion of intracellular immunoglobulin or in an unbound form, but it was not secreted except in assocmtion with polymeric immunoglobulins. In sections of human lymphoid tissues, J chain was found in the majority of cells that contained immunoglobulins (5).

Parallel synthesis of immunoglobulins and J chain in pokeweed mitogen- stimulated normal cells and in lymphoblastoid cell lines
The synthesis of intracellular J chains was found to be closely associated with that of intracellular immunoglobulin, regardless of its class, during the process of B-cell differentiation. This parallelism between the synthesis of J chain and immunoglobulin was particularly evident in their coincident appearance in serial observations of pokeweed mitogen (PWM)-stimulated lymphocytes. The intens...
متن کاملProduction of predominantly polymeric IgA by human peripheral blood lymphocytes stimulated in vitro with mitogens
Human peripheral blood lymphocytes (PBL) were cultured for various time periods (up to 8 d) in the presence of pokeweed mitogen (PWM), lipopolysaccharide, or Epstein-Barr virus. Cell-free supernates were fractionated on a standardized ultrogel AcA 22 column and the proportion of polymeric and monomeric IgA was determined by radioimmunoassay. The results demonstrate that PBL stimulated with thes...
متن کاملProduction of granulocyte-macrophage colony-stimulating factor by Abelson virus-induced tumorigenic mast cell lines.
We have recently described a system that supports the development of continuously growing and tumorigenic cell lines after infection of individual multilineage hematopoietic colonies with Abelson murine leukemia virus (A-MuLV). We now provide definitive evidence that these transformed lines express features characteristic of mast cells. Although these lines have been maintained in some cases fo...
متن کاملPolyclonal activation of human peripheral blood B lymphocytes by formaldehyde-fixed Salmonella paratyphi B. I. Immunoglobulin production without DNA synthesis
A "new" polyclonal activator of human peripheral blood B cells, formaldehyde-fixed Salmonella paratyphi B, is described. This bacterium does not stimulate cell proliferation as measured by incorporation of tritiated thymidine but does stimulate a subpopulation of B cells to secrete large amounts of IgM, IgG, and IgA in 7-day cell cultures. The immunoglobulins (Ig) produced by cells responding t...
متن کاملAutoantibodies to B lymphocytes in a patient with hypoimmunoglobulinemia. Characterization and pathogenic role.
In a young woman with ulcerative colitis, hypoimmunoglobulinemia, and humoral immunodeficiency, lymphocyte counts vary between 600 and 1,000 per mm(3) with 0.5-1.5% bone marrow-derived (B) cells and 98-99% thymus-derived (T) cells. Anti-lymphocyte antibodies were detected by immunofluorescence and by microlymphocytotoxicity with increased reactivity at +4 degrees C. They belonged to the IgM cla...
